Karakterisering van immuuncel-afgeleide extracellulaire vesicles en het bestuderen van functionele impact op celmilieu

Het onderhavige rapport belicht chronologische eisen voor extracellulaire vesicle (EV) isolatie van microglia of bloedmacrofagen. Microglia-afgeleide EV's werden geëvalueerd als regelgevers van de neurite uitgroei, terwijl bloed macrofaag-afgeleide EV's werden bestudeerd in de controle van C6 glioom cel invasie in in vitro testen. Het doel is om deze EV-functies als immuunbemiddelaars in specifieke micro-omgevingen beter te begrijpen.

Abstract

De neuro-inflammatoire toestand van het centrale zenuwstelsel (CNS) speelt een belangrijke rol in fysiologische en pathologische omstandigheden. Microglia, de inwonende immuuncellen in de hersenen, en soms de infiltrerende beenmerg-afgeleide macrofagen (BMDMs), reguleren het ontstekingsprofiel van hun micro-omgeving in het CNS. Men aanvaardt nu dat de extracellulaire vesicle (EV) populaties uit immuuncellen fungeren als immuunbemiddelaars. Zo zijn hun verzameling en isolatie belangrijk om hun inhoud te identificeren, maar ook hun biologische effecten op ontvangende cellen te evalueren. De huidige gegevens benadrukken chronologische vereisten voor EV-isolatie van microgliacellen of bloedmacrofagen, waaronder de ultracentrifugatie en grootte-uitsluiting chromatografie (SEC) stappen. Een niet-gerichte proteomische analyse liet de validatie van eiwithandtekeningen als EV-markers toe en karakteriseerde de biologisch actieve EV-inhoud. Microglia-afgeleide EV's werden ook functioneel gebruikt op de primaire cultuur van neuronen om hun belang als immuunbemiddelaars in de neurite uitgroei te beoordelen. De resultaten toonden aan dat microglia-afgeleide EV's bijdragen aan het vergemakkelijken van de neurite uitgroei in vitro. Tegelijkertijd werden bloed macrofaag-afgeleide EV's functioneel gebruikt als immuunbemiddelaars in sferoïde culturen van C6 glioomcellen, de resultaten waaruit blijkt dat deze EV's de gliomacelinvasie in vitro controleren. Dit rapport belicht de mogelijkheid om de EV-gemedieerde immuuncelfuncties te evalueren, maar ook de moleculaire basis van een dergelijke communicatie te begrijpen. Deze ontcijfering kan het gebruik van natuurlijke vikjes en/of de in vitro voorbereiding van therapeutische vikjes bevorderen om immuuneigenschappen in de micro-omgeving van CNS-pathologieën na te bootsen.

Veel neuropathologieën zijn gerelateerd aan de neuro-inflammatoire toestand die een complex mechanisme is dat steeds meer wordt overwogen, maar nog steeds slecht begrepen omdat de immuunprocessen divers zijn en afhankelijk zijn van de celomgeving. De CNS-aandoeningen hebben immers niet systematisch dezelfde activeringssignalen en immuuncelpopulaties en dus zijn de pro- of ontstekingsremmende reacties moeilijk te beoordelen als oorzaken of gevolgen van pathologieën. De hersenen resident macrofagen genaamd "microglia" lijken te zijn op de interface tussen het zenuwstelsel en het immuunsysteem1. Microglia hebben een myeloïde oorsprong en zijn afge....

    1. Kweek commerciële rat primaire microglia (2 x 106 cellen) (zie de tabel van materialen) in Dulbecco's gemodificeerde Eagle medium (DMEM) aangevuld met 10% exosoomvrij serum, 100 U/mL penicilline, 100 μg/mL streptomycin en 9,0 g/L bij 37 °C en 5% CO2.
    2. Verzamel het geconditioneerde medium na een cultuur van 48 uur en ga door naar de isolatie van EV's.

Een van de belangrijkste uitdagingen voor het toeschrijven van biologische effecten aan extracellulaire vesicles (EV's) is de mogelijkheid om de EV's te isoleren van het hele cultuurmedium. In dit rapport presenteren we een methode met behulp van ultracentrifugatie (UC) en size-exclusion chromatografie (SEC) die is gekoppeld aan de grootschalige analyse van eiwithandtekeningen om EV-markers te valideren en bioactieve verbindingen te identificeren.
