Dit artikel beschrijft een workflow van technieken die worden gebruikt voor het testen van nieuwe kandidaat-mediatoren van melanoommetastase en hun werkingsmechanisme (s).

Abstract

Metastase is een complex proces waarbij cellen barrières moeten overwinnen die slechts onvolledig worden gemodelleerd door in vitro assays. Een systematische workflow werd opgezet met behulp van robuuste, reproduceerbare in vivo modellen en gestandaardiseerde methoden om nieuwe spelers in melanoommetastase te identificeren. Deze benadering maakt het mogelijk om gegevens in specifieke experimentele stadia te laten afleiden om de rol van een gen in metastase nauwkeurig te karakteriseren. Modellen worden vastgesteld door genetisch gemodificeerde melanoomcellen via intracardiale, intradermale of subcutane injecties in muizen te introduceren, gevolgd door monitoring met seriële in vivo beeldvorming. Zodra vooraf vastgestelde eindpunten zijn bereikt, worden primaire tumoren en/of metastasen-dragende organen geoogst en verwerkt voor verschillende analyses. Tumorcellen kunnen worden gesorteerd en onderworpen aan een van de verschillende 'omics'-platforms, waaronder eencellige RNA-sequencing. Organen ondergaan beeldvorming en immunohistopathologische analyses om de totale last van metastasen te kwantificeren en hun specifieke anatomische locatie in kaart te brengen. Deze geoptimaliseerde pijplijn, inclusief gestandaardiseerde protocollen voor engraftment, monitoring, weefseloogst, verwerking en analyse, kan worden gebruikt voor patiënt-afgeleide, kortetermijnculturen en gevestigde menselijke en muizencellijnen van verschillende vaste kankertypen.

De hoge mortaliteit geassocieerd met gemetastaseerd melanoom in combinatie met een toenemende incidentie van melanoom wereldwijd1 (een geschatte toename van 7,86% tegen 2025) vraagt om nieuwe behandelingsbenaderingen. Vooruitgang in doelontdekking hangt af van reproduceerbare modellen van metastase, een zeer complex proces. Gedurende de stappen van de gemetastaseerde cascade moeten melanoomcellen talloze barrières overwinnen om ontwijking van het immuunsysteem en kolonisatie van verre weefsels te bereiken2. OPMERKING: De dierprocedures die betrokken zijn bij het volgende protocol zijn goedgekeurd door de New York University Institutional Animal Care and Use Committee (IACUC). Alle procedures worden uitgevoerd in faciliteiten die zijn goedgekeurd door de Association for Assessment and Accreditation of Laboratory Animal Care International (AAALAC). Het doel van dit technisch rapport is om een gestandaardiseerde, top-to-bottom workflow te bieden voor het onderzoek naar potentiële actoren in melanoommetastase. Omdat in vivo experimenten kostbaar en tijdrovend kunnen zijn, zijn strategieën om de efficiëntie te maximaliseren en de waarde van de verkregen informatie te verhogen van het grootste belang.

Het is absoluut noodzakelijk om overal complementaire benaderingen te gebruiken om bevindingen binnen hetzelfde experiment te crossva.......
