Alkaline Extraction

Alkaline extraction is a biochemical method that uses high-pH conditions to release and separate target molecules from cells or complex sample materials. The alkaline solution changes the ionization of biomolecules and disrupts noncovalent interactions; in plasmid DNA preparation, sodium hydroxide and detergent lyse cells and denature chromosomal DNA and proteins, while subsequent neutralization promotes plasmid DNA renaturation and precipitation of cellular debris. This approach provides a relatively simple way to isolate plasmid DNA for cloning, sequencing, and other molecular biology applications, while illustrating how pH-dependent chemical properties can drive biomolecular separation.

The alkaline comet assay measures DNA strand breaks in eukaryotic cells. By adding an Endonuclease III or human 8-oxoguanine-DNA-N-glycosylase digestion step, the assay can efficiently detect oxidative DNA damage. We describe methods for using these assays to detect DNA damage in rat liver.
