Oxygen Uptake Analysis

Oxygen uptake analysis is the measurement of how quickly cells, tissues, organisms, or biochemical systems consume oxygen, providing an indicator of aerobic respiration and metabolic activity. In respirometry, oxygen concentration or partial pressure is monitored over time in a sealed or controlled chamber; the rate of decline reflects electron transport and oxidative phosphorylation when temperature, substrate availability, and other conditions are controlled. In biochemistry, this approach helps assess mitochondrial function, respiratory efficiency, enzyme or drug effects, and energy metabolism. Comparing oxygen uptake under different substrates or inhibitors can reveal defects in respiration and clarify how biological systems produce and use energy.

Synaptosomal dopamine uptake and high-performance liquid chromatography analysis represent experimental tools to investigate dopamine homeostasis in mice by assessing the function of the dopamine transporter and levels of dopamine in striatal tissue, respectively. Here we present protocols to measure dopamine tissue content and assess the functionality of the dopamine transporter.

This video demonstrates an in vitro technique for estimating amino acid uptake by bone marrow-derived stromal cells. Na+-dependent amino acid transporters mediate the uptake of radiolabeled amino acids inside the cells, and the cellular uptake is estimated by measuring the radioactivity.

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
