Cell Cycle Sorting

Cell cycle sorting is a flow cytometry-based method that separates cells according to their position in the cell cycle, enabling phase-specific biochemical and molecular analysis. Cells are labeled with a fluorescent DNA-binding dye, and the cytometer measures DNA content as individual cells pass through a laser; computational gates distinguish G0/G1, S, and G2/M populations before charged droplets direct selected cells into separate collection tubes. This approach produces enriched cell-cycle fractions for studying DNA replication, protein regulation, gene expression, and cell-cycle responses to drugs or other experimental conditions.

We report a Fluorescent Activated Cell Sorting (FACS)-based method to isolate neural stem cells (NSCs) and their progeny from the subventricular zone (SVZ) of the adult mouse brain. Applied to Fluorescence Ubiquitination Cell Cycle Indicator (FUCCI) transgenic mice, it allows the study of cell cycle progression by live imaging.

A method for isolating specific cell types from plant material is demonstrated. This technique employs transgenic marker lines expressing fluorescent proteins in particular cell types, cellular dissociation and Fluorescence Activated Cell Sorting. Additionally, a growth setup is established here that facilitates treatment of Arabidopsis thaliana seedlings prior to cell sorting.

Flow Cytometry-Based Cell Sorting for Different Circulatory B Cell Populations

In this video, we describe a protocol for the flow cytometry-based sorting of naïve B cell, memory B cell, and plasmablast plasma cell subpopulations from human peripheral blood B cells. The different B cell subpopulations can be distinguished based on fluorophore-based antibody binding to the CD19, CD27, and CD38 receptors expressed on the B cell surfaces, followed by cell sorting.
