Peptide Affinity Reagents

Peptide affinity reagents are short, designed or selected peptide molecules that bind specific biomolecular targets, providing versatile tools for biochemical detection, purification, and analysis. Their activity depends on selective noncovalent interactions, including hydrogen bonding, electrostatic forces, hydrophobic contacts, and shape complementarity, which can produce stable peptide–target complexes under defined conditions. Researchers can immobilize these reagents on solid supports to capture proteins or other analytes, or attach labels for detection in assays. Because peptides are compact and readily modified, peptide affinity reagents support studies of protein interactions, biomarker measurement, drug discovery, and the development of targeted diagnostic or purification methods.

Biopanning bacterial display libraries is a proven technique for discovery of peptide affinity reagents, a robust alternative to antibodies. The semi-automated sorting method herein has streamlined biopanning to decrease the occurrence of false positives. Here we illustrate the thought process and techniques applied in evaluating candidates and minimizing downstream analysis.
