Peptide Array Synthesis

Peptide array synthesis is a high-throughput method for producing many spatially addressable peptides on a shared solid support, enabling systematic analysis of sequence-dependent molecular interactions. Typically, amino acids are added through repeated cycles of protected residue coupling and deprotection, either at defined positions on a membrane or on a chemically functionalized surface; each location records a distinct peptide sequence. In biochemistry, these arrays help map antibody or receptor-binding sites, identify enzyme substrates, profile post-translational modification recognition, and evaluate sequence specificity. Their parallel format reduces sample use and accelerates screening, although signals may depend on peptide length, surface attachment, and assay conditions.

Personalized Peptide Arrays for Detection of HLA Alloantibodies in Organ Transplantation

Mismatches in human leukocyte antigen (HLA) sequences between organ donor and recipient pairs are the major cause of antibody-mediated rejection in organ transplantation. Here we present the use of custom antigen arrays that are based on individual donors' HLA sequences to probe anti-donor HLA alloantibodies in organ recipients.

Peptide tertiary amides (PTAs) are a superfamily of peptidomimetics that include but are not limited to peptides, peptoids and N-methylated peptides. Here we describe a synthetic method which combines both split-and-pool and sub-monomer strategies to synthesize a one-bead one-compound library of PTAs.

This video demonstrates an assay to detect anti-donor HLA alloantibodies in organ recipients. A peptide array is synthesized to represent potential epitopes for antibody-mediated organ rejection. The array is incubated with the recipient's serum, containing alloantibodies that bind to their target epitopes. Antibody binding is detected using chemiluminescence to identify donor-specific HLA epitopes that incite alloantibody reactivity.
