Spectrophotometric Confirmation

Spectrophotometric confirmation is a method that uses light absorption to verify the identity, purity, or concentration of a biochemical sample. A spectrophotometer measures how much light a sample absorbs at selected wavelengths, and the resulting absorbance spectrum can be compared with an expected profile or interpreted using the Beer-Lambert law to relate absorbance to concentration. In biochemistry, this approach helps confirm proteins, nucleic acids, enzymes, and other analytes after isolation or preparation. It provides a rapid, non-destructive assessment that supports quality control, experimental validation, and the interpretation of downstream biochemical measurements.

Spectrophotometric Determination of an Equilibrium Constant

2023

Source: Laboratory of Dr. Michael Evans — Georgia Institute of Technology The equilibrium constant, K, for a chemical system is the ratio of product concentrations to reactant concentrations at equilibrium, each raised to the power of their respective stoichiometric coefficients. Measurement of K involves determination of these concentrations for systems in chemical equilibrium. Reaction systems containing a single colored component can be studied spectrophotometrically. The relation between...

Confirmation Biases

2020

The confirmation bias is the tendency to focus on information that confirms our existing beliefs and ignore information that is inconsistent with our expectations. For example, if you think that your professor is not very nice, you notice all of the instances of rude behavior exhibited by the professor while ignoring the countless pleasant interactions he is involved in on a daily basis. Have you ever fallen prey to the confirmation bias, either as the source or target of such bias? Another...

Humane Euthanasia and Confirmation of Death in Rodents

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Humane endpoints in rodents are predefined clinical, behavioral, or physiological indicators that signal an animal is experiencing significant pain, distress, or declining health. When these signs are observed, timely...

2020

Glutathione S-transferases (GSTs) are detoxification enzymes involved in the metabolism of numerous chemotherapeutic drugs. Overexpression of GSTs is correlated with cancer chemotherapy resistance. One way to counter this phenotype is to use inhibitors. This protocol describes a method using a spectrophotometric assay to screen for potential GST inhibitors.

Spectrophotometric Assay to Determine Glycogen Synthase Activity
