In Vivo Collagen Remodeling

In vivo collagen remodeling is the continual restructuring of collagen-rich extracellular matrix within living tissues, a process that maintains mechanical integrity while enabling growth, repair, and adaptation. Fibroblasts synthesize and organize new collagen, while matrix metalloproteinases and other proteases cleave existing fibers; cross-linking, cellular forces, and tissue mechanics then shape the matrix’s composition and architecture. In bioengineering, understanding these coordinated changes helps researchers design biomaterials and engineered tissues that integrate with host tissue, regulate wound healing, and restore function. Quantifying collagen turnover and alignment can also clarify mechanisms of fibrosis, regeneration, and disease progression.

Imaging Denatured Collagen Strands In vivo and Ex vivo via Photo-triggered Hybridization of Caged Collagen Mimetic Peptides

This procedure demonstrates in vivo near IR fluorescence imaging of collagen remodeling activities in mice as well as ex vivo staining of collagens in tissue sections using caged collagen mimetic peptides that can be photo-triggered to hybridize with denatured collagen strands.
