Perivascular Cell Recruitment

Perivascular cell recruitment is the directed movement and association of support cells, such as pericytes and vascular smooth muscle cells, with blood vessels, a process that helps establish and maintain vascular stability. In response to signals from endothelial cells, injured tissue, or biomaterials, perivascular cells migrate toward developing vessels, adhere to the vessel surface, and contribute extracellular matrix and regulatory signals. In bioengineering, controlling this process supports the formation of stable, functional microvascular networks in engineered tissues, organoids, and regenerative scaffolds. Understanding recruitment mechanisms can improve vascular integration, tissue perfusion, and the design of therapies for repair and regeneration.

Use of Human Perivascular Stem Cells for Bone Regeneration

Human perivascular stem cells (PSCs) are a novel stem cell class for skeletal tissue regeneration similar to mesenchymal stem cells (MSCs). PSCs can be isolated by FACS (fluorescence activated cell sorting) from adipose tissue procured during standard liposuction procedures, then combined with an osteoinductive scaffold to achieve bone formation in vivo.

Isolation of Perivascular Multipotent Precursor Cell Populations from Human Cardiac Tissue

Human cardiac tissue harbours multipotent perivascular precursor cell populations that may be suitable for myocardial regeneration. The technique described here allows for the simultaneous isolation and purification of two multipotent stromal cell populations associated with native blood vessels, i.e. CD146+CD34- pericytes and CD34+CD146- adventitial cells, from the human myocardium.

The video demonstrates a method for investigating the impact of stromal cells on leukocyte recruitment. In a parallel plate flow chamber model, pro-inflammatory cytokines induce the expression of adhesion receptors by endothelial cells, facilitating neutrophil recruitment and migration. The cross-talk between stromal cells and endothelial cells diminishes neutrophil recruitment to the endothelial cells.
