Photoacoustic Wave Detection

Photoacoustic wave detection is the measurement of ultrasound waves generated when modulated or pulsed light is absorbed by a material, linking optical contrast with acoustic sensing. In bioengineering, short laser pulses deposit energy in tissue or a target chromophore; rapid thermoelastic expansion produces pressure waves that transducers detect and convert into signals related to absorber distribution and concentration. This approach supports noninvasive imaging of blood vessels, oxygenation, pigments, and engineered materials, combining the molecular specificity of optical methods with the depth and spatial resolution of ultrasound. Understanding detector response, signal processing, and light–tissue interactions is essential for developing photoacoustic instruments for diagnostics, monitoring, and biomedical research.

Photoacoustic cystography (PAC) has a great potential to map urinary bladders, a radiation sensitive internal organ in pediatric patients, without using any ionizing radiation or toxic contrast agent. Here we demonstrate the use of PAC for mapping urinary bladders with an injection of optical-opaque tracers in rats in vivo.

In this video, we present a photoacoustic flow cytometric detection of ovarian cancer cells bearing nanoparticles in a circulating in vitro flow chamber. This non-invasive technique successfully identifies circulating tumor cells in a flow system utilizing a contrast agent.
