Specimen Staining

Specimen staining is a laboratory technique that uses dyes or labels to increase contrast and reveal the structure, composition, or distribution of components within biological specimens. Stains work through selective interactions with cellular molecules or tissue features, often depending on chemical properties such as charge, affinity, or permeability, allowing targeted structures to appear differently under microscopy. In bioengineering, specimen staining supports the assessment of cells, tissues, biomaterials, and engineered constructs by showing morphology, organization, viability, or specific molecular markers. These visual results help researchers evaluate fabrication methods, cell-material interactions, tissue development, and disease-related changes.

The zebrafish embryo is an excellent model for developmental biology research. During embryogenesis, zebrafish develop with a yolk mass, which presents three-dimensional challenges for sample observation and analysis. This protocol describes how to create two-dimensional flat mount preparations of whole mount in situ (WISH) stained zebrafish embryo specimens.

We developed protocols and designed a custom apparatus to enable embedding of millimeter-scale specimens. We present sample preparation procedures with an emphasis on embedding in acrylic resin and polyimide tubing to achieve rigid immobilization and long-term storage of specimens for the interrogation of tissue architecture and cell morphology by micro-CT.

Isolating electrical and thermal effects on electrically assisted deformation (EAD) is very difficult using macroscopic samples. Metallic sample micro- and nanostructures together with a custom test procedure have been developed to evaluate the impact of applied current on the formation without joule heating and evolution of dislocations on these samples.
