Spycatcher Spytag

SpyCatcher–SpyTag is a protein-ligation system that enables researchers to connect proteins and other biological components through a specific, irreversible covalent bond. A short SpyTag peptide binds its engineered partner, SpyCatcher, which catalyzes formation of an internal isopeptide bond between reactive amino acid side chains without requiring additional enzymes or chemical reagents. In bioengineering, this modular interaction supports protein purification, surface immobilization, biomolecular assembly, and construction of vaccines, biosensors, and self-assembling materials. Because the partners can be genetically fused to diverse targets and function under many physiological conditions, the system provides a flexible strategy for designing stable protein architectures and studying complex biological functions.

This video demonstrates a method to generate multivalent-displaying outer membrane vesicle (OMV) vaccines. Mammalian cells are transfected to express and secrete viral antigens fused to a SpyTag and a polyhistidine tag. These antigens are then purified using a nickel-nitrilotriacetic acid (Ni-NTA) affinity column.
