Tlr4 Lps Interaction

The TLR4-LPS interaction is the molecular recognition process by which Toll-like receptor 4 detects bacterial lipopolysaccharide, a major component of Gram-negative cell walls, and initiates innate immune signaling. In this pathway, LPS is transferred by lipopolysaccharide-binding protein and CD14 to the MD-2–TLR4 complex, promoting receptor dimerization and activation of MyD88- and TRIF-dependent signaling that regulates inflammatory gene expression. In bioengineering, understanding this interaction supports the design of endotoxin detection assays, immune-responsive biomaterials, and drug-delivery systems with controlled inflammatory effects. It also helps researchers evaluate biocompatibility and reduce harmful immune activation in engineered devices.

This protocol is based on LPS and ATP-induced death of PMA-differentiated THP-1 macrophages. We use flow cytometry to analyze Annexin V and 7-AAD double staining to detect cell death, using the whole cell and employing scanning electron microscopy to observe the cell membrane morphology.
