Calcium Green-5n Fluorescence

Calcium Green-5N fluorescence is a calcium-imaging method that uses a low-affinity fluorescent indicator to detect changes in free calcium concentration, particularly in environments with high calcium levels. The dye increases its fluorescence when calcium binds to its chelating site, allowing the signal intensity to reflect calcium availability under appropriate calibration conditions. In biological techniques, researchers use Calcium Green-5N to monitor calcium storage and release in organelles, cells, and membrane systems, including rapid intracellular signaling events. Its low affinity helps extend measurements into calcium ranges that higher-affinity indicators may not accurately resolve, supporting studies of calcium homeostasis, transport, and physiology.

A method for fluorescent staining of fixed biological material with the specific DNA label methyl green is described. Methyl green is used in a diluted aqueous solution and is very resistant to photobleaching. Its far-red emission allows for deep specimen imaging, making it particularly adequate for whole embryos.
