Contact Mode Afm

Contact mode AFM is an atomic force microscopy technique that images surfaces by maintaining continuous contact between a sharp probe and the sample, enabling nanoscale measurements of biological structures. As the probe scans across the surface, interactions between its tip and the sample cause cantilever deflection; a feedback system adjusts the probe height to maintain a defined force and records surface topography. In biology, contact mode AFM can characterize cell membranes, biomolecular assemblies, and other soft or rigid specimens with high spatial resolution. These measurements reveal surface organization, mechanical features, and structural changes relevant to cell biology, microbiology, and biomaterials research.

The lack of tools to measure material properties and tensional parameters in vivo prevents validating their roles during development. We employed atomic force microscopy (AFM) and nanoparticle-tracking to quantify mechanical features on the intact zebrafish embryo yolk cell during epiboly. These methods are reliable and widely applicable avoiding intrusive interventions.

To obtain basic information on the sorption and recycling of gold from aqueous systems the interaction of Au(III) and Au(0) nanoparticles on S-layer proteins were investigated. The sorption of protein polymers was investigated by ICP-MS and that of proteinaceous monolayers by QCM-D. Subsequent AFM enables the imaging of the nanostructures.
