Nascent Rna Visualization

Nascent RNA visualization is a set of methods used to detect and track newly synthesized RNA in cells, revealing when and where transcription occurs. These approaches typically label RNA during a defined pulse of transcription, using metabolically incorporated nucleotide analogs or sequence-specific probes, and then detect the labeled transcripts through fluorescence or chemical reactions. In biology, nascent RNA visualization helps measure transcriptional activity, distinguish newly produced transcripts from preexisting RNA, and examine RNA processing and localization. The technique supports studies of gene regulation, developmental changes, cellular responses, and disease-associated alterations in transcription.

Trophoblast giant cells (TGCs) play a key role in the placenta to ensure a healthy pregnancy. We present a protocol for assessing the transcriptional status of genes in TGCs by nascent fluorescent in situ hybridization on cryostat sections of post-implantation embryos or short-term cultures of embryonic day 7 ectoplacental cones.

Isolation of Ribosome Bound Nascent Polypeptides in vitro to Identify Translational Pause Sites Along mRNA

A technique to identify translational pause sites on mRNA is described. This procedure is based on isolation of nascent polypeptides accumulating on ribosomes during in vitro translation of a target mRNA, followed by the size analysis of the nascent chains using a denaturing gel electrophoresis.
