Nitric Oxide Detection

Nitric oxide detection is the measurement of nitric oxide (NO), a short-lived signaling molecule that regulates vascular tone, neurotransmission, and immune activity in biological systems. Detection methods exploit NO’s rapid reactions with oxygen, superoxide, and heme centers, or measure its electrochemical oxidation; fluorescent probes can respond when NO-derived products modify a reporter molecule, while analytical assays quantify its stable products, nitrite and nitrate. In biology, these approaches help map NO production in cells and tissues, clarify signaling pathways, and evaluate changes associated with inflammation, cardiovascular dysfunction, neuronal activity, and therapeutic responses.

In this article we describe an adapted relatively easy method using the fluorescence dye diaminofluorescein-2 diacetate (DAF-2DA) and dihydroethidium (DHE) for en face simultaneous detection and visualization of intracellular nitric oxide (NO) and superoxide anion (O2.−) respectively, in freshly isolated intact aortas of an obesity mouse model.

The endogenous production of nitric oxide (NO) regulates a wide variety of biological functions. It is becoming increasingly clear that disruption or dysregulation of NO based signaling is involved in many human diseases. Methods to quantify relevant NO metabolites may provide novel diagnostic or prognostic biomarkers for human disease.

Electron paramagnetic resonance (EPR) spectroscopy was employed to detect nitric oxide from bovine aortic endothelial cells and superoxide radical anion from human neutrophils using iron (II)-N-methyl-D-glucamine dithiocarbamate, Fe(MGD)2 and 5,5-dimethyl-1-pyroroline-N-oxide, DMPO, respectively.

Levels of the inflammatory cell-signaling molecule nitric oxide (NO) are commonly assayed using Griess reagent. In this protocol, we have created a modified Griess assay utilizing live Drosophila brain tissue in order to detect the secretion of NO in a simple, quantifiable and highly repeatable method.

This video demonstrates the precise technique for inducing a white matter stroke in an anesthetized mouse by injecting a nitric oxide inhibitor at targeted brain coordinates within the white matter. This leads to blood vessel constriction and inhibition of blood supply, resulting in neuronal damage and a brain stroke.
