Protein Isolation

Protein isolation is the process of separating a specific protein from cells, tissues, or biological mixtures so its structure and function can be studied. It typically begins with cell lysis, followed by clarification and purification steps that exploit differences in solubility, charge, size, or binding affinity; methods such as precipitation and chromatography progressively remove contaminants. In biology, isolated proteins support enzyme activity assays, structural analysis, antibody production, diagnostics, and biotechnology research. The quality and purity of the preparation directly affect experimental reliability, making careful control of buffers, temperature, and processing conditions essential for preserving protein stability and biological activity.

Source: Dembowski, J. A., et al. Purification of Viral DNA for the Identification of Associated Viral and Cellular Proteins. J. Vis. Exp. (2017).This video demonstrates the isolation of protein–viral DNA complexes from cell nuclei using click chemistry and magnetic bead purification. The protocol involves labeling viral genomes with alkyne-modified nucleotides, biotinylation via a click reaction, and binding to streptavidin-coated magnetic beads. Following washing and heat elution, the purified...

A tool and chemistries are described to sequentially isolate nucleic acids followed by proteins from a sample without the need for electricity. The tool consists of a sorbent held within a transfer pipette while the isolation chemistries are based on solid-phase extraction principles. The isolated macromolecules can be analyzed by immuno-based and PCR-based assays.

Novel RNA-Binding Proteins Isolation by the RaPID Methodology
