Stromal Cell-derived Inducing Activity

Stromal cell-derived inducing activity is the capacity of certain stromal cells to promote neural differentiation in pluripotent stem cells, making it a useful model for studying how cellular environments regulate fate. In co-culture systems, stromal cells provide instructive signals through direct cell contact and released factors that shift stem cells toward neural progenitor and neuronal lineages. Researchers use this activity to investigate early nervous-system development, identify signals controlling lineage commitment, and generate neural cells for disease modeling and regenerative research. Because the responsible signals may involve multiple interacting components, the system also supports studies of cell-based induction mechanisms.

Described here is a method for producing exogenous cytokine in patient-derived xenograft (PDX) mice via weekly intraperitoneal injection of a cytokine-transduced stromal cell line. This method broadens the utility of PDX and provides the option for transient or sustained exogenous cytokine delivery in a multitude of PDX models.

Adipose-derived stromal cells may be useful for engineering new tissue from a patient's own cells. We present a protocol for the isolation of a subpopulation of human adipose-derived stromal cells (ASCs) with increased osteogenic potential, followed by application of the cells in an in vivo calvarial healing assay.
