Hcl Decalcification

HCl decalcification is a tissue-processing technique that removes mineralized calcium salts from bone and other calcified specimens, making them pliable enough for sectioning and microscopic examination. Hydrochloric acid dissolves calcium phosphate by lowering the solution’s pH, but exposure time and acid concentration must be controlled because prolonged treatment can impair tissue morphology and reduce the quality of nucleic acids or protein signals. In cancer research, the method supports histopathological analysis of bone tumors, tumor metastases, and calcified biopsy samples, helping researchers assess cellular architecture, disease progression, and treatment responses while balancing rapid processing against preservation of molecular detail.

Calcified Artery Preparation and Processing with Preserved Morphology and RNA for Digital Spatial Profiling

2026

This protocol includes a step-by-step workflow for calcified vascular specimens: tissue handling, decalcification, RNA validation, calcification level detection, and region-of-interest selection strategies on the Nanostring GeoMx Digital Spatial Profiler (DSP). The goal is to present a comprehensive method for preserving vascular tissue morphology and RNA for reliable spatial transcriptomic analysis.

This includes measuring the mass or volume of reactants and products, methods of heating and cooling reaction mixtures, proper selection and handling of glassware, and separating solids and liquids by vacuum filtration. First, you'll synthesize benzoic acid by protonating...

A Cell-Based Therapy for Autoimmune Arthritis in a Murine Model

2025

This video showcases a cell-based therapy for autoimmune arthritis in mice. iPSC-derived T regulatory cells expressing ovalbumin-specific receptors are injected with methylated bovine serum albumin (mBSA) emulsified in an adjuvant. Murine antigen-presenting cells (APCs) process and present the mBSA to T cells, prompting T cell activation and migration to the knee joint's synovial membrane. Subsequent injections involve mBSA alone in the left knee and a combination of mBSA and ovalbumin in the...

Tissue Preparation and Immunostaining of Mouse Sensory Nerve Fibers Innervating Skin and Limb Bones

Immunocytochemical identification of peripheral sensory nerve fiber subtypes (and detection of protein expression therein) are key to the understanding of molecular mechanisms underlying peripheral sensation. Here we describe methods for preparation of peripheral/visceral tissue samples, such as skin and limb bones, for specific immunostaining of peripheral sensory nerve fibers.
