Hypoxia-sensitive Car

A hypoxia-sensitive CAR is an engineered chimeric antigen receptor designed to regulate immune-cell activity according to local oxygen levels, an important strategy for targeting oxygen-poor tumor regions. In cancer research, hypoxia-responsive elements can control CAR transcription, while oxygen-dependent degradation domains can promote receptor breakdown under normoxia and stabilize it when oxygen is scarce, linking receptor availability to the tumor microenvironment. This conditional design may focus CAR T-cell recognition and cytotoxicity within hypoxic tumors, potentially improving tumor selectivity while reducing activity in healthy, well-oxygenated tissues. It also provides a platform for studying how microenvironmental conditions influence cellular immunotherapies.

We introduce a novel hypoxic chamber system for use with aquatic organisms such as frog and zebrafish embryos. Our system is simple, robust, cost-effective and allows the induction and sustainment of hypoxia in vivo and for up to 48 h. We present 2 reproducible methods to monitor the effectiveness of hypoxia.
