Open Inguinal Surgery

Open inguinal surgery is a conventional operative approach that uses an incision in the groin to access the inguinal canal, lymph nodes, or nearby tissues, making it important for treating and evaluating disease in this region. During the procedure, the surgeon separates tissue layers under direct visualization, identifies relevant vessels and nerves, and may remove abnormal tissue or inguinal lymph nodes for pathological examination. In cancer research and care, this approach supports regional staging, detection of metastatic spread, and treatment planning for malignancies that involve the groin. Surgical specimens can also provide tissue for molecular and translational studies.

Intravital Microscopy of the Inguinal Lymph Node

A technique for performing intravital microscopy of the inguinal lymph node (LN) is outlined. Such technique allows for real-time, in vivo study of the lymph node microvasculature and structure both during homeostasis and infection. This technique can be adapted to cell trafficking studies and to other lymph node sites.

In this video, we demonstrate the technique to image the inguinal lymph node in a mouse model using ultra-high-frequency ultrasound imaging. This non-invasive imaging technique allows the monitoring of the change in lymph node volume in the animal over time.

Harvesting an Inguinal Draining Lymph Node from a Murine Model

Incision of the abdominal region provides access to the inguinal lymph node, which is surgically harvested. The harvested translucent inguinal lymph node can be used for immunological studies.

This protocol describes the method of determining basal and forskolin-stimulated lipolysis in inguinal fat pads obtained from normal chow diet (NCD) or high fat diet (HFD) ± capsaicin fed wild type mice. As an index for lipolysis, glycerol release was measured from inguinal adipose fat pads.
