Transposon-based Transgenesis

Transposon-based transgenesis is a genetic engineering method that uses mobile DNA elements to introduce and stably express foreign genes in a cell or organism, making it useful for studying cancer biology. A transposase enzyme recognizes terminal sequences flanking a transgene, excises the transposon from a donor construct, and inserts it into genomic DNA, where the cargo can be inherited and expressed. In cancer research, engineered transposons support insertional mutagenesis screens that activate oncogenes or disrupt tumor-suppressor genes, helping identify pathways that drive tumor development. The approach also enables reproducible cancer models and functional testing of candidate genes, although insertion site and copy number can influence results.
