Cell Suspension Seeding

Cell suspension seeding is a cell culture method in which dispersed cells are introduced into a growth vessel or scaffold at a defined density, enabling controlled initiation of an experiment. Cells are suspended in culture medium, gently mixed to promote uniform distribution, and transferred to the culture surface, where suitable conditions support attachment, survival, and proliferation. In cancer research, this approach helps establish reproducible tumor cell cultures, compare growth behaviors, and prepare cells for assays of drug response, invasion, or cell signaling. Consistent seeding density improves experimental comparability and supports the development of models for studying tumor biology and therapeutic effects.

This video describes the technique of seeding mesenchymal stem cells (MSCs) onto a biodegradable poly(lactic acid) (PLA) scaffold. The prepared scaffolds can be implanted into the surgical resection cavity of the brain from where they may migrate towards tumor foci. Further, these cells can be engineered to express therapeutic proteins to stimulate apoptosis in cancer cells.

Measurement of Vacuolar and Cytosolic pH In Vivo in Yeast Cell Suspensions

Vacuolar and cytosolic pH can be measured in live yeast (S. cerevisiae) cells using ratiometric fluorescent dyes localized to specific cellular compartments. We describe procedures for measuring vacuolar pH with BCECF-AM, which localizes to the vacuole in yeast, and cytosolic pH with a cytosolic ratiometric pH-sensitive GFP (yeast pHluorin).

The tail-suspension test is validated as an experimental procedure to assess antidepressant efficacy of drug treatments in mice. Mice are suspended by their tails for six minutes and escape-related behaviors are assessed. We describe procedures used in conducting the tail suspension test.
