Alkaline Conditions

Alkaline conditions are chemical environments with a pH above 7, typically created by dissolved bases, and they strongly influence reaction behavior, solubility, and molecular charge. In water, bases accept protons or generate hydroxide ions, shifting acid–base equilibria; the resulting hydroxide concentration and pH can alter nucleophile availability, ionization states, and the stability of compounds. Chemists use alkaline conditions to control synthesis, hydrolysis, precipitation, extraction, and analytical measurements, while materials and environmental studies use them to evaluate corrosion, mineral transformations, and pollutant mobility. Understanding these effects helps researchers select suitable reagents and predict reaction outcomes.

The alkaline comet assay measures DNA strand breaks in eukaryotic cells. By adding an Endonuclease III or human 8-oxoguanine-DNA-N-glycosylase digestion step, the assay can efficiently detect oxidative DNA damage. We describe methods for using these assays to detect DNA damage in rat liver.
