Pta Submonomer

Pta submonomer is a specialized chemical building block used to introduce a defined functional group during the stepwise assembly of peptoids and related oligomers. In the submonomer method, an activated haloacetic acid first acylates the growing chain, and a subsequent nucleophilic substitution by an amine replaces the leaving group, installing the Pta-derived side chain. Repeating these two reactions enables precise control over sequence and composition while avoiding the need to prepare and isolate each complete monomer in advance. This modular strategy supports the synthesis of peptoid libraries for studying molecular recognition, biomolecular interactions, materials properties, and structure–function relationships.

A simple and general manual peptoid synthesis method involving basic equipment and commercially available reagents is outlined, enabling peptoids to be easily synthesized in most laboratories. The synthesis, purification and characterization of an amphiphilic peptoid 36mer is described, as well as its self-assembly into highly-ordered nanosheets.

Peptide tertiary amides (PTAs) are a superfamily of peptidomimetics that include but are not limited to peptides, peptoids and N-methylated peptides. Here we describe a synthetic method which combines both split-and-pool and sub-monomer strategies to synthesize a one-bead one-compound library of PTAs.

An Efficient Method for the Synthesis of Peptoids with Mixed Lysine-type/Arginine-type Monomers and Evaluation of Their Anti-leishmanial Activity

A protocol to synthesize peptoids with mixed cationic functionality in the same sequence is presented (lysine- and arginine-type monomers). Subsequent testing of these compounds against Leishmania mexicana, the protozoan parasites that cause cutaneous leishmaniasis, is also described.

This article describes a process for tuning the size and crosslinking density of covalently crosslinked nanoparticles from linear polyesters containing pendant functionality. By tailoring synthesis parameters (polymer molecular weight, pendant functionality incorporation, and crosslinker equivalents), a desired nanoparticle size and crosslinking density can be achieved for drug delivery applications.
