Synthetic Protein Engineering

Synthetic protein engineering is the design and production of proteins with tailored sequences, structures, and functions, making it an important approach in modern chemistry and biotechnology. Researchers modify DNA sequences through rational design, gene synthesis, or directed evolution, then express the resulting proteins and evaluate how amino acid substitutions alter folding, stability, binding, or catalytic activity. These methods support the creation of improved enzymes for chemical synthesis, proteins that selectively recognize molecular targets, and engineered materials with specialized properties. By linking molecular structure to function, synthetic protein engineering advances biocatalysis, therapeutic development, biomaterials research, and the study of complex chemical processes.

Engineering Adherent Bacteria by Creating a Single Synthetic Curli Operon

The design of a synthetic operon encoding both the secretory apparatus and the structural monomers of curli fibers is described. Overproduction of these amyloids and adherent polymers allows a measurable gain of adherence of the E. coli chassis1. Easy ways to visualize and quantify adherence are explained.

Protein transduction enables the direct delivery of biologically active proteins into cells. In contrast to conventional methods such as DNA transfection or viral transduction this non-invasive paradigm allows highly efficient cellular manipulation in a titratable manner circumventing cellular toxicity and the risk of oncogenic transformation by permanent genetic modification.

We demonstrate the basic technique to molecularly engineer and evolve synthetic Adeno-associated viral (AAV) gene therapy vectors via DNA family shuffling. Moreover, we provide general guidelines and representative examples for selection and analysis of individual chimeric capsids with enhanced properties on target cells in culture or in mice.
