Blastomere Injection

Blastomere injection is a micromanipulation technique that introduces genetic material, molecular probes, or other reagents into an individual cell of a cleavage-stage embryo to study early development. Under microscopic control, a fine glass micropipette penetrates the embryo and delivers a controlled volume into the blastomere, allowing investigators to alter gene activity or label specific cell lineages. In developmental biology, the method helps reveal how early cells divide, communicate, and acquire distinct fates, while supporting analyses of gene function, lineage allocation, mosaicism, and embryo patterning. Results from these experiments can clarify mechanisms that shape normal development and developmental defects.

After neural tube formation, the neuroepithelium constricts and folds while the tube fills with embryonic cerebrospinal fluid (eCSF) to form the embryonic brain ventricles. We developed this ventricle injection technique to better visualize the fluid filled space in contrast to the neuroepithelial shape in a live embryo.
