Neuroblast Division

Neuroblast division is the process by which neural progenitor cells multiply and generate the diverse neurons and glial cells that form the nervous system, making it central to brain development. In many developing tissues, neuroblasts divide asymmetrically: coordinated spindle orientation, cell polarity, and unequal inheritance of fate determinants produce one self-renewing progenitor and one differentiating daughter cell. Studying this process reveals how progenitor pools are maintained, how neuronal identity and tissue architecture emerge, and how errors in division can disrupt neural development. Experimental analyses of neuroblast lineages support research on cell fate, neurogenesis, and mechanisms underlying neurological disease.

This protocol details a streamlined method used to conduct live cell imaging in the context of an intact larval brain. Live cell imaging approaches are invaluable for the study of asymmetric neural stem cell divisions as well as other neurogenic and developmental processes, consistently uncovering mechanisms that were previously overlooked.

2013

Neuroblast migration is a crucial step in postnatal neurogenesis. The protocol described here can be used to investigate the role of candidate regulators of neuroblast migration by employing DNA/small hairpin RNA (shRNA) nucleofection and a 3D migration assay with neuroblasts isolated from the rodent postnatal rostral migratory stream.

Cell Division - Concepts

Cell division is fundamental to all living organisms and required for growth and development. As an essential means of reproduction for all living things, cell division allows organisms to transfer their genetic material to their offspring. For a unicellular organism, cellular division generates a completely new organism. For multicellular organisms, cellular division produces new cells for the general development of the organism, as well as produces healthy cells to replace damaged cells from...

Observing the Cell Cycle in a Root Tip ExpandHypotheses: The experimental hypothesis is that in root tips slices that have been treated with nocodazole, a chemical that interferes with microtubular polymerization, all of the cells will be arrested at the same stage of the cell cycle and that in untreated onion tip slices all of the different stages of the cell cycle will be visualized. The null hypothesis is that there will be no differences in mitosis between the two groups and that different...
