Centrifugal Microfluidics

Centrifugal microfluidics is a lab-on-a-disc approach that uses rotation to control tiny liquid volumes in integrated channels, enabling compact analysis with minimal reagent use. During operation, centrifugal pressure drives fluid radially outward, while channel geometry, rotational speed, capillary valves, and metering chambers regulate timing, volume, mixing, and separation. In engineering, these systems combine fluid handling and assay steps on disposable discs, supporting automated sample preparation and diagnostic testing in portable platforms. Their simple actuation and scalable design are useful for point-of-care devices, environmental analysis, and research on integrated microsystems.

Here, we demonstrate a simple production method for size-controllable, monodisperse, water-in-oil (W/O) microdroplets using a capillary-based centrifugal microfluidic device. This method requires only a small sample volume and enables high-yield production. We expect this method will be useful for rapid biochemical and cellular analyses.

This protocol demonstrates how to achieve femto molar detection sensitivity of proteins in 10 µL of whole blood within 30 min. This can be achieved by using electrospun nanofibrous mats integrated in a lab-on-a-disc, which offers high surface area as well as effective mixing and washing for enhanced signal-to-noise ratio.

Working with Centrifuges

Source: Robert M. Rioux & Suprita Jharimune, Pennsylvania State University, University Park, PA Earth's gravitational force is capable of separating heterogeneous mixtures. However, many systems avert this type of separation owing to the length of time usually required in such cases. Centrifugation is one of the most powerful tools for the separation of heterogeneous mixtures1. It involves the application of centripetal force for sedimentation of the heavier phase, leading to separation of...

Centrifugation

Centrifugation is a separation technique based on differences in density or size. It is commonly used to separate solids from aqueous interferents. During centrifugation, the sample is placed in centrifugation tubes and spun at high angular velocity, which allows centrifugal force to act differentially on the different densities or masses of the components. After spinning, the supernatant liquid is decanted. Depending on the specific application, either the pellet or the supernatant is retained...
