Pressure Gradient Analysis

Pressure Gradient Analysis is the study of how pressure changes across distance, time, or a system, providing a basis for understanding and controlling fluid behavior in engineering. By calculating the pressure gradient as the spatial rate of pressure change, engineers can predict flow direction, driving force, and pressure losses in relation to fluid properties, geometry, and boundary conditions. This analysis supports the design and evaluation of pipelines, pumps, porous materials, ventilation systems, and aerodynamic components. It also helps identify bottlenecks, optimize energy use, assess system performance, and diagnose abnormal flow or leakage in engineered systems.

The laser heated diamond anvil cell combined with synchrotron micro-diffraction techniques allows researchers to explore the nature and properties of new phases of matter at extreme pressure and temperature (PT) conditions. Heterogeneous samples can be characterized in situ under high pressure by 2D mapping and combined powder, single-crystal and multigrain diffraction approaches.

Density Gradient Ultracentrifugation

2023

Density gradient ultracentrifugation is a common technique used to isolate and purify biomolecules and cell structures. This technique exploits the fact that, in suspension, particles that are more dense than the solvent will sediment, while those that are less dense will float. A high-speed ultracentrifuge is used to accelerate this process in order to separate biomolecules within a density gradient, which can be established by layering liquids of decreasing density in a centrifuge tube. The...

The manuscript here provides a simple set of methods for analysing the secretion and diffusion of fluorescently tagged ligands in Xenopus. This provides a context for testing the ability of other proteins to modify ligand distribution and allowing experiments that may give insight into mechanisms regulating morphogen gradients.
