Sgrna Target Selection

sgRNA target selection is the process of choosing a guide RNA sequence that directs CRISPR-associated nucleases to a specific genomic site. Selection relies on sequence complementarity between the sgRNA and target DNA, the presence of a compatible protospacer-adjacent motif (PAM), and evaluation of nearby sequence features that may influence cleavage efficiency and off-target activity. In genetics, carefully designed guides enable targeted gene disruption, sequence modification, or regulation of gene expression. Comparing candidate sites and minimizing potential matches elsewhere in the genome improves editing specificity, experimental reliability, and the interpretation of phenotypic results.

Nanoblade-Based Delivery of Nucleic Acid Cargo: A Technique to Deliver Cas9-sgRNA Complex to Target Cells via Virus-Like Particles

2025

This video demonstrates a technique for nanoblade-based delivery of Cas9-sgRNA ribonucleoprotein complex inside target cells for genome editing. Nanoblades are virus-like particles devoid of the ability to multiply and infect neighboring cells, therefore are very useful for rapid and dose-dependent transportation of bio- and nanomaterials.

Ethics in Target Audience Selection

2024

Ethics in targeting audiences is a crucial aspect of marketing and business practices. It involves understanding and respecting the rights, interests, and dignity of consumers while conducting any promotional activities or communications. Unethical targeting can lead to exploitation, manipulation, or harm, particularly for vulnerable groups like children, older people, or those with low financial literacy. Ethical targeting respects consumer privacy, avoids intrusive advertising, and ensures...

Using Phage Display to Select Proteins with High Affinity to a Target Protein

In this video, we demonstrate the phage display technique to isolate pre-engineered phages expressing proteins with high affinity to the target protein.

Simultaneous Eye Tracking and Single-Neuron Recording to Identify Target-Selective Neurons

2025

The video demonstrates an assay to identify target-selective neurons via simultaneous eye-tracking and single-neuron recordings. A human participant is tasked with identifying a target object among an array of objects in a display, and the eye movement is tracked. Simultaneously, single neurons' firing rate is recorded via electrodes implanted in the brain. The data from both recordings are correlated to assess the presence of target-selective neurons.

Targeting Gray Rami Communicantes in Selective Chemical Lumbar Sympathectomy

We present a protocol for selective chemical lumbar sympathectomy (CLS), which inactivates only gray rami communicantes and not the sympathetic trunk. Selective CLS can help achieve therapeutic efficacy in vasodilation, sweat reduction, and pain relief that are comparable to conventional CLS, and serious complications, particularly ureteropelvic damages, can be reduced.
