Egg Decontamination

Egg decontamination is the controlled removal or inactivation of microorganisms on an eggshell before the egg is used in infection, immunology, or virology research. The process typically combines surface cleaning with a validated disinfectant and controlled contact time, followed by aseptic handling, to reduce microbial entry through shell pores without damaging the embryo or compromising experimental procedures. In embryonated chicken eggs, decontamination helps prevent bacterial and fungal contamination during viral inoculation, propagation, and harvesting. Reliable shell disinfection improves reproducibility, protects cultures from confounding organisms, and supports studies of pathogen growth, host responses, vaccine production, and infection mechanisms.

Decontamination for Laboratory Biosafety

2023

Robert M. Rioux and Zhifeng Chen, Pennsylvania State University, University Park, PA Decontamination is essential for laboratory biosafety, as the accumulation of microbial contamination in the laboratory can lead to the transmission of disease. The degree of decontamination can be classified as either disinfection or sterilization. Disinfection aims to eliminate all pathogenic microorganisms, with the exception of bacterial spores on lab surfaces or equipment. Sterilization, on the other hand,...

Egg Laying Assay: A Method to Quantify the Egg-Laying Behavior of C. elegans

2023

C. elegans egg-laying behavior can provide important information in developmental, reproductive, and anatomical studies. This video introduces the important steps of an egg laying assay and shows a sample experiment in which a drug is tested for reproductive toxicity.

Drosophila Egg Collection and Dechorionation: A Method to Remove the Outermost Egg Layer

2023

The Drosophila embryo is surrounded by protective outer membranes. To gain access to the embryo, the chorion–the outermost of the membranes–is often removed. This video describes a commonly used method of dechorionation using sodium hypochlorite. The example protocol demonstrates the procedure while employing sterile techniques necessary for axenic or gnotobiotic animal rearing.

Egg Windowing: A Method of Opening Fertilized Chicken Egg Shell to Expose Chorioallantoic Membrane Vasculature
