Immunoreactivity Assessment

Immunoreactivity assessment is the evaluation of how strongly a biological sample reacts with a specific antigen or antibody, providing evidence of immune recognition and helping characterize immune responses. In practice, assays expose serum, cells, or tissue to a defined target and detect antigen-antibody binding or immune-cell activation through measurable signals such as color, fluorescence, or protein bands. These measurements support detection of pathogen exposure, evaluation of vaccine-induced immunity, characterization of autoimmune reactivity, and validation of diagnostic reagents. In immunology and infection research, comparing signal intensity with controls helps distinguish specific responses from background reactivity and improves interpretation of disease-associated immune markers.

Here we present a protocol to visualize spatial correlation of calcitonin gene-related peptide (CGRP)-immunoreactive nerve fibers and blood vessels in the cranial dura mater using immunofluorescence and fluorescent histochemistry with CGRP and phalloidin, respectively. In addition, the origin of these nerve fibers was retrograde traced with a fluorescent neural tracer.

We developed a protocol to assess well-being in mice during procedures using general anesthesia. A series of behavioral parameters indicating levels of well-being as well as glucocorticoid metabolites were analyzed. The protocol can serve as a general aid to estimate the degree of severity in a scientific, animal-centered manner.
