Mouse Serum Analysis

Mouse serum analysis is the laboratory assessment of soluble molecules in mouse blood serum to characterize immune status, inflammation, or responses to infection. After blood is allowed to clot and cells are removed, analytical assays such as immunoassays measure targets including antibodies, cytokines, complement proteins, or pathogen-specific antigens. Comparing serum profiles between experimental groups can reveal immune activation, exposure, disease progression, or treatment effects. In immunology and infection research, these measurements complement cellular and tissue analyses, helping connect systemic molecular changes with host defense and support evaluation of vaccines, therapeutics, and disease models.

This protocol describes in detail the method for generating neural progenitors from embryonic stem cells using a serum-free monolayer method. Serum utilized in embryo cultures contains unknown components that can affect the outcome of experiments especially in studies involving signaling interactions. Here we utilized a serum-free oxygenated culture system and show that mid-gestation mouse embryos cultured for 16-40 hr exhibit morphological development comparable to embryos developing in utero.

The proposed work assesses the diagnostic potential of direct and amplified surface plasmon resonance imaging (SPRi) assays, particularly for the detection of recombinant human growth hormone in spiked human serum, by comparing SPRi results directly with commercially available enzyme-linked immunosorbent assay (ELISA) kit.
