Virus Quantification

Virus quantification is the measurement of viral abundance or infectious activity in a sample, an essential step in studying infection, immunity, and antiviral responses. Depending on the objective, researchers may count infectious particles with plaque or focus-forming assays, which detect infected-cell zones produced through viral replication, or measure viral genomes using quantitative PCR. These approaches provide complementary information because genome copies do not necessarily represent infectious virions. In immunology and infection research, virus quantification helps characterize replication kinetics, compare pathogen strains, evaluate vaccine and antiviral efficacy, and relate viral burden to disease severity or host immune responses.

This protocol permits the isolation of Epstein-Barr virus particles from the human P3HR1 cell line upon inducing the viral lytic cycle with phorbol 12-myristate 13-acetate. DNA is subsequently extracted from the viral preparation and subjected to real-time PCR to quantify the viral particle concentration.
