Cell Membrane Integrity

Cell membrane integrity is the ability of a cell’s plasma membrane to maintain its structure, selective permeability, and internal environment, making it essential for normal cellular function and survival. The phospholipid bilayer, membrane proteins, and associated cholesterol regulate movement of ions and molecules while preserving electrochemical gradients; physical, chemical, or biological injury can disrupt this organization, causing leakage, altered signaling, and loss of homeostasis. In medicine, assessing membrane integrity helps identify cellular damage, distinguish viable from nonviable cells, and evaluate disease processes, toxic effects, and responses to treatments. Fluorescent permeability assays and measurements of released intracellular components provide practical indicators of membrane injury.

In this video, we demonstrate a fluorescence-detection size-exclusion chromatography technique to screen for green fluorescent protein (GFP)-fused membrane protein stability in Escherichia coli following detergent solubilization. Proteins exhibiting a symmetrical peak in the size-exclusion profile with little or no free GFP indicate purified proteins with minimum degradation and aggregation, which can subsequently be selected for structure-function analysis.
