Heterotopic Cervical Model

A heterotopic cervical model is an experimental transplantation model in which a donor organ, most commonly a heart, is implanted in the recipient’s neck while the native organ remains in place. The graft is connected to cervical blood vessels through vascular anastomoses, allowing it to receive circulation without replacing the recipient’s original organ; investigators can then assess graft activity and survival. In medicine, this model supports studies of transplant rejection, ischemia-reperfusion injury, immunosuppressive therapies, and graft function. Its accessible location and separation from essential native-organ function make it useful for controlled investigation of transplantation biology and treatment strategies.

The murine cervical heart transplantation model is well suited for immunological as well as ischemia reperfusion injury studies. We modified the procedure using a non-suture cuff technique and performed more than 1,000 successful transplants with this approach. Herein, we provide additional details of this technique to supplement the video.

In the present protocol, a mouse heart transplantation model is used for investigating the mechanism of cardiac allograft rejection. In this heterotopic heart transplantation model, operation efficiency is improved, and the survival of cardiac grafts is ensured by a cervical end-to-end anastomosis of heart implantation using a modified Cuff technique.
