Reagent Loading Procedure

Reagent loading procedure is a controlled method for placing chemical or biological reagents into an assay, cartridge, instrument, or reaction vessel so they are ready for analysis or treatment-related testing. It typically involves identifying each reagent, measuring or dispensing the required volume, following a defined loading order, and maintaining suitable conditions to prevent contamination, degradation, or unintended reactions. In medicine, consistent reagent loading supports reliable diagnostic assays, laboratory workflows, and pharmaceutical research by improving reaction reproducibility and reducing handling errors. Standardized procedures also help laboratories compare results, maintain quality control, and adapt testing platforms for routine or automated use.

Preparing Anhydrous Reagents and Equipment

Source: Laboratory of Dr. Dana Lashley - College of William and Mary Demonstrated by: Timothy Beck and Lucas Arney Many reactions in organic chemistry are moisture-sensitive and must be carried out under careful exclusion of water. In these cases the reagents have a high affinity to react with water from the atmosphere and if left exposed the desired reaction will not take place or give poor yields, because the reactants are chemically altered. In order to prevent undesired reactions with H2O...

EDTA: Auxiliary Complexing Reagents

EDTA titrations are usually carried out in highly basic conditions, where the fully deprotonated form of EDTA, Y4−, actively complexes with the free metal ions in the solution. Several metal ions precipitate as hydrous oxide (hydroxides, oxides, or oxyhydroxides) under these conditions, lowering the concentration of free metal ions in the solution. For this reason, auxiliary complexing agents or ligands such as ammonia, tartrate, citrate, or triethanolamine are used in EDTA titrations to...

Acid Halides to Ketones: Gilman Reagent

2025

Lithium dialkyl cuprate, also known as Gilman reagents, selectively reduces acid halides to ketones. The acid chloride is treated with Gilman reagent at −78 °C in the presence of ether solution to produce a ketone in good yield. As shown below, the mechanism proceeds in two steps. First, one of the alkyl groups of the reagent acts as a nucleophile and attacks the acyl carbon of the acid chloride to form a tetrahedral intermediate. This is followed by the reformation of the carbon–oxygen double...
