Structure Preservation

Structure preservation is the maintenance of a specimen’s original anatomical, cellular, or tissue organization so that it remains suitable for medical examination and interpretation. In pathology and biomedical research, this usually depends on prompt fixation, controlled processing, and appropriate storage: chemical fixatives stabilize cellular components and inhibit autolysis and microbial degradation, while careful handling limits distortion, shrinkage, or mechanical damage. Preserved structure supports accurate histological staining, microscopy, and diagnosis from biopsy or surgical specimens, and helps researchers relate tissue architecture to disease mechanisms, treatment response, and clinical outcomes. Consistent preservation also improves comparability among specimens and strengthens the reliability of medical findings.

A simple procedure of vibratome sectioning the organ of Corti, followed by immunohistochemistry and confocal microscopy is described. This procedure allows for improved preservation of the fine cytoarchitecture of the mammalian organ of Corti, and consequently allows for accurate quantification of cell types.
