Brain Tissue Preservation

Brain tissue preservation is the set of methods used to maintain the structure, molecular components, and cellular information of brain samples after collection. Chemical fixation stabilizes tissue by crosslinking proteins, while cryoprotection and controlled freezing help limit ice-crystal damage and preserve fragile structures during storage or sectioning. In neuroscience, these approaches support histology, immunohistochemistry, microscopy, and molecular analyses of neural circuits, disease-related changes, and developmental processes. Selecting a preservation method that matches the intended analysis is essential because processing conditions can affect tissue morphology, antigen accessibility, and nucleic acid quality.

Source: Kirchner, K.N., et al., A Hydrophobic Tissue Clearing Method for Rat Brain Tissue. J. Vis. Exp. (2020).This video demonstrates the method of hydrophobic tissue clearing of a rat brain tissue sample. The sample is sequentially dehydrated in methanol, treated with a dichloromethane and methanol mixture to dissolve lipids and pigments, and transferred to dibenzyl ether for complete tissue clearing, enabling detailed imaging without thin sectioning.
