Longitudinal Muscle

Longitudinal muscle is muscle tissue organized with fibers running parallel to the long axis of a body segment or organ, enabling controlled shortening along that axis. In neuroscience, its activity links neural commands to movement and offers a tractable system for studying neuromuscular control. When motor neurons release neurotransmitter at neuromuscular junctions, intracellular calcium rises in muscle fibers, allowing actin-myosin interactions to generate force and contraction. These muscles contribute to locomotion, posture, and organ movement, while their anatomy and physiology help researchers analyze motor circuits, muscle coordination, and disorders that impair movement.

Assessing the Structural Integrity of the Dorsal Longitudinal Muscle Neuromuscular Junctions in Drosophila

2025

This video demonstrates a protocol for assessing the structural integrity of neuromuscular junctions in the dorsal longitudinal muscles of Drosophila. The procedure involves staining neurons and muscles with two distinct antibodies and comparing fluorescence patterns between mutant and wild-type flies. A reduction in neuronal staining in mutant flies indicates neurodegeneration, while the muscle structure remains intact.

Sometimes we want to see how people change over time, as in studies of human development and lifespan. When we test the same group of individuals repeatedly over an extended period of time, we are conducting longitudinal research. Longitudinal research is a research design in which data-gathering is administered repeatedly over an extended period of time. For example, we may survey a group of individuals about their dietary habits at age 20, retest them a decade later at age 30, and then again...

Longitudinal studies are also widely used in other medical and social science fields. For instance, in cardiovascular research, they can monitor patients' health over decades to identify risk factors for heart disease, such as high cholesterol or smoking, and evaluate the long-term effectiveness of preventive measures. Similarly, in mental health studies, researchers might follow individuals from adolescence into adulthood to understand the development and progression of conditions like...

C. elegans Muscle Area Measurements: A Standardized Method to Quantify Striated Muscle Morphology

2023

C. elegans have two types of muscles, single sarcomere and multiple sarcomere, or striated muscles. This video describes the arrangement of the striated body wall muscles, which are responsible for a worm’s locomotion and includes a sample protocol to measure muscle morphology using image analysis software.
