Nmda Stimulus

An NMDA stimulus is an experimental activation of N-methyl-D-aspartate receptors, glutamate-gated ion channels that help regulate excitatory signaling and synaptic plasticity in the nervous system. NMDA receptor opening requires ligand binding and sufficient membrane depolarization to relieve the magnesium block, allowing calcium and other cations to enter the neuron. In neuroscience, controlled NMDA stimulation is used to investigate long-term potentiation, neuronal excitability, calcium-dependent signaling, and excitotoxicity, including how altered receptor activity may contribute to neurological disease. These studies clarify mechanisms of learning, memory, development, and synaptic injury.

Here, we describe the use of a genetically directed alkaline phosphatase (AP) labeling method to investigate the anatomical features and phenotypes of mouse retinas under NMDA-induced excitotoxicity. In conjunction with several retinal ganglion cell (RGC) subtype-specific reporter lines, we uncover novel phenotypes in dying RGCs throughout the degenerative process.

Inducing Axonal Varicosities with a Micromechanical Stimulus on Neurons

This video demonstrates the induction of axonal varicosities by applying micromechanical stimulus on neurons. The system comprises a micropipette attached to a syringe containing buffer via tubing. Using a micromanipulator, the micropipette is positioned above the neurons. Fluid pressure is then applied to induce varicosities in axons.
