Oxygen Concentration Measurement

Oxygen concentration measurement is the quantitative determination of dissolved or gaseous oxygen in a sample, tissue, or experimental environment, providing a key readout of cellular metabolism and physiological state. In common optical methods, an oxygen-sensitive luminophore emits light whose intensity or lifetime decreases when molecular oxygen collides with and quenches its excited state; calibration converts this signal into oxygen concentration. In neuroscience, these measurements help characterize oxygen delivery, consumption, and hypoxic stress in brain tissue, neural cultures, and model systems. Combined with electrophysiology, imaging, or metabolic assays, oxygen data can link neural activity to energy demand and reveal changes associated with disease or injury.

Cerebral Blood Oxygenation Measurement Based on Oxygen-dependent Quenching of Phosphorescence

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.
