Retinal Whole Mount

Retinal whole mount is a tissue-preparation technique in which the retina is dissected from the eye, flattened, and mounted as an intact sheet to preserve its two-dimensional organization. After fixation, radial cuts help spread the curved tissue, allowing researchers to label retinal cells, synapses, blood vessels, or disease-related changes with histochemical or immunofluorescent methods. Microscopy of the preparation reveals spatial patterns across the retinal surface, including cell density, neuronal distribution, vascular architecture, and lesion extent. In neuroscience, retinal whole mounts support studies of visual circuitry, development, neurodegeneration, regeneration, and the effects of injury or experimental treatments.
