Tyrosine Hydroxylase Immunostaining

Tyrosine hydroxylase immunostaining is an antibody-based method used to visualize cells and fibers that express tyrosine hydroxylase, the rate-limiting enzyme in catecholamine synthesis. In fixed nervous tissue, primary antibodies bind tyrosine hydroxylase, and labeled secondary antibodies produce a fluorescent or chromogenic signal that reveals the distribution and morphology of immunoreactive structures. In neuroscience, this technique helps identify dopaminergic and noradrenergic neurons, map their projections, and assess changes in catecholaminergic systems after injury, disease, development, or experimental treatment. Image-based analysis can further support comparisons of cell number, staining intensity, and fiber density across brain regions or conditions.

Receptor Tyrosine Kinases

2025

Receptor tyrosine kinases or RTKs are membrane-bound receptors that phosphorylate specific tyrosine on protein substrates. RTKs regulate cellular growth, differentiation, survival, and migration. They contain an extracellular ligand binding domain, a transmembrane domain, and a cytosolic tail with intrinsic kinase activity. Several extracellular signaling molecules activate RTKs in one or more ways and relay the signal downstream. Ligands such as platelet-derived growth factor (PDGF) or...

Immunostaining of Cells Encapsulated in a 3D Hydrogel System

2025

This video demonstrates a technique for immunostaining cells encapsulated in a 3D hydrogel. The hydrogel culture containing neural progenitor cells is fixed, permeabilized, and treated with a blocking solution to prevent non-specific immunostaining. A primary antibody cocktail is added to bind specific proteins, followed by fluorescently labeled secondary antibodies and a nuclear stain. The sample is then mounted on a slide and observed under a confocal microscope.
