Lipid Water Partition Coefficient

Lipid-water partition coefficient is a measure of how a chemical compound distributes between a lipid phase and an aqueous phase, providing an indicator of its relative lipophilicity and behavior in biological systems. It is determined by allowing the compound to equilibrate between immiscible phases and calculating the concentration ratio; pH and ionization can strongly influence the result, especially for weak acids and bases. In pharmacology, this property helps predict membrane permeation, gastrointestinal absorption, tissue distribution, and whether a drug remains in aqueous fluids or partitions into lipid-rich environments. Alongside solubility and protein-binding data, it supports drug design, formulation, and interpretation of pharmacokinetic behavior.

A technique to probe the lipid raft partitioning of fluorescent proteins at the plasma membrane of living cells is described. It takes advantage of the disparity in diffusion times of proteins located inside or outside of lipid rafts. Acquisition can be performed dynamically in control conditions or after drug addition.
