Methacholine Carbachol Bethanechol

Methacholine, carbachol, and bethanechol are direct-acting muscarinic cholinergic agonists that mimic acetylcholine and produce parasympathetic effects in pharmacology. By binding muscarinic receptors, they activate intracellular signaling pathways that alter smooth-muscle tone and glandular secretion, although their clinical actions and receptor selectivity differ. Methacholine is primarily used in bronchial challenge testing, where inhalation-induced airway narrowing helps assess bronchial hyperresponsiveness. Carbachol is used to produce miosis and reduce intraocular pressure, while bethanechol stimulates bladder and gastrointestinal smooth muscle to support urinary or intestinal motility; together, these drugs illustrate how muscarinic receptor activation can be applied diagnostically and therapeutically.

We describe a method for generating Precision-cut Lung Slices (PCLS) and immunostaining them to visualize the localization of various immune cell types in the lung. Our protocol can be extended to visualize the location and function of many different cell types under a variety of conditions.
