Chlamydia pneumoniae are pathogenic bacteria that can infect the human brain.

To detect these bacteria, take a deparaffinized, rehydrated, bacteria-infected human brain section with exposed protein antigens and inactivated endogenous phosphatase.

Add a blocking solution to prevent non-specific antibody binding.

Incubate with primary antibodies that bind with the target bacterial antigen.

Wash with a buffer to remove unbound antibodies.

Add the blocking solution to increase the specificity of the secondary antibody.

Incubate with alkaline phosphatase-conjugated secondary antibodies, which bind to primary antibodies.

Rinse with water to remove excess antibodies.

Add a chromogenic substrate that reacts with alkaline phosphatase, forming a colored precipitate.

Rinse with water to remove excess precipitate.

Add a dye to stain nuclei.

Add mounting media to mount the section.

Under a light microscope, visualize stained bacteria in the brain tissue.
